Protocol Synopsis Review.
LiveDocument ReviewIs this Phase 2 protocol synopsis feasible, internally consistent and safe to take to site selection?
Protocol Synopsis Review is a ready-made use case for Clinical Operations. It ships under the Document Review template.
For clinical operations leads. You get an item-by-item ledger with verdicts and risk, the substantiation each claim needs, the required changes and what the review could not check. Claims are cited or marked as gaps.

The decision
Is this Phase 2 protocol synopsis feasible, internally consistent and safe to take to site selection?
Who reviews
The SPIRIT statement lists the items a clinical trial protocol should address. The seats use it as a checklist for completeness and add their own lane’s checks.
| Seat | What it reviews | What it works from |
|---|---|---|
| Clinical Operations Director | Feasibility: eligibility, visit burden, sites and enrollment. | Site and enrollment data. |
| Head of Pharmacovigilance | Safety monitoring, stopping rules and reporting. | Safety data for the compound or class. |
| Director Quality Assurance | Consistency between sections and readiness for site-level execution.2 | ICH E8(R1) on quality by design. |
| Regulatory Strategist | Whether the design will support the intended claim and the agency’s expectations.2 | ICH guidance and agency feedback. |
What the review does
Consistency across sections. The ledger checks that objectives, endpoints, schedule of assessments and statistics agree, and lists each mismatch with its location.1
Completeness against SPIRIT. A synopsis is short by nature, so the review notes what a full protocol must add and does not penalize the synopsis for it.1
Quality built in. The quality seat asks whether the design focuses on what matters to participant safety and the reliability of the result.2
Recommended length
Standard
Four lanes bear on this document. Standard depth gives each a seat, the ledger and the substantiation table in about four pages.
You can pick another length on the form. The other two stay available.
Assumptions the review makes
The review states what it assumes about the document and the material supplied, and says what each gap limits. Examples for this document:
- The synopsis reflects the current draft.
- Sites have the capacity the schedule assumes.
- The endpoint has been discussed with the agency.

Framework
The decision practice behind the memo itself is on the template page.
What gets flagged
Any mismatch between objectives and endpoints, any visit schedule the sites cannot deliver, and any safety rule left undefined.
What arrives
- The document under review
- Overall recommendation
- The review, by seat
- Item-by-item ledger
- Substantiation and evidence gaps
- Required changes and conditions for approval
- Rebuttal round
- What this review could not check
What it will not do
This is analyst judgment from AI specialists and not a regulatory, medical or legal sign-off. It cannot check original data or the approved label, and the decision stays with you.
It does not rewrite the protocol.
It is analyst judgment from AI specialists and not a regulatory, medical or legal sign-off.
It does not rewrite the document. It states the change and leaves the writing to the author.
It does not check original data or the approved label held by the company. Those checks need a real reviewer.

About the Panel service
Each panelist answers in their own voice, then the discussion is drawn together. You get the question and who was in the room, every position in full, what they agreed on, where they split, and what follows. Disagreements are written down rather than smoothed into a consensus nobody actually holds, because the split is usually the part you needed.
